Image Scanning Microscopy to Investigate Polycomb Protein Colocalization onto Chromatin

Super-resolution microscopy has been recently applied to understand the 3D topology of chromatin at an intermediated genomic scale (kilobases a few megabases), as this corresponds sub-diffraction spatial crucial for regulation gene transcription. In context, polycomb proteins are very renowned repressors that organize into multiprotein complexes Polycomb Repressor Complex 1 (PRC1) and 2 (PRC2). PRC1 PRC2 operate onto according complex mechanism, which was recapitulated working model. Here, we present functional colocalization study 100–140 nm resolution targeting well histone mark H3K27me3 by Image Scanning Microscopy (ISM). ISM offers more flexible alternative diffraction-unlimited SRMs such STORM STED, it is perfectly suited investigate mesoscale PRC assembly. Our data suggest partially simultaneous effort in locally shaping topology.
